A new study suggests the effectiveness of Viagra can be enhanced by boosting levels of brain-derived neurotrophic factor (BDNF) in a mouse model of Alzheimer's disease. The study, by Dr. Susan O'Brien, of University of Texas M. D. Anderson and colleagues, was published today in the.
The findings, published in the journal Alzheimer's Research, are being analyzed by several groups of scientists at the University of Texas M. Anderson School of Medicine at Baylor College of Medicine in Houston.
In the study, researchers studied the effects of three different treatments — sildenafil (Viagra), tadalafil (Cialis) and vardenafil (Levitra) on brain-derived neurotrophic factor (BDNF) levels in mice: sildenafil (Viagra), tadalafil (Cialis) and vardenafil (Levitra). To measure the effects of these treatments, they were crossed to the mouse model of Alzheimer's disease and treated with the drugs sildenafil (Viagra) and tadalafil (Cialis). These experiments were designed to study the effectiveness of these drugs on BDNF levels in mice.
"Our study suggests that these drugs can enhance the effectiveness of the drugs that help to treat Alzheimer's disease," said Dr. O'Brien, from the Department of Neurology at Baylor College of Medicine and the co-author of the study. "This is important because the clinical benefit of drugs in the treatment of Alzheimer's disease is less than the benefit of medications in the treatment of Alzheimer's disease, particularly drugs that are not approved by the FDA."
In addition, the researchers were also interested in testing whether sildenafil, tadalafil and vardenafil were able to help patients suffering from Alzheimer's disease.
The drugs in the study were sildenafil (Viagra) and tadalafil (Cialis) and were administered for up to 12 weeks before the animals were observed for any signs of disease. The researchers found that sildenafil, tadalafil and vardenafil improved brain-derived neurotrophic factor levels by up to a mean of 4.5 percent in these animals.
The drugs were effective in treating Alzheimer's disease in the animals in the study, but they had no effect on the levels of BDNF in the mice.
Dr. O'Brien and colleagues used the mouse model to study the effects of these drugs on the brains of Alzheimer's disease patients. They studied 17 Alzheimer's patients who were treated with the drugs sildenafil, tadalafil and vardenafil. All patients were on the drugs for at least one year.
Results showed that the treatment with sildenafil, tadalafil and vardenafil increased BDNF levels in the mice by 60 percent. However, those levels were reduced by up to 90 percent in the treated mice.
In addition, the levels of BDNF were elevated in the treated mice by 60 percent and 40 percent in the untreated group, respectively.
The researchers also found that the drugs reduced the levels of total BDNF, as well as the levels of phosphorylated forms of BDNF, by about 80 percent in the treated mice.
These results were supported by data showing that sildenafil, tadalafil and vardenafil improved the levels of total and phosphorylated forms of BDNF in the brain of Alzheimer's patients.
The research was published today in the journal Alzheimer's Research. The scientists, led by Dr. Susan O'Brien, are the first researchers to study the effects of these drugs on levels of BDNF in the brain of Alzheimer's patients. The researchers were funded by the National Institute of Neurological Disorders and Stroke, National Institute of Health, the National Institutes of Health and the National Institute of Child Health and Human Development.
"Our findings show that sildenafil, tadalafil and vardenafil are effective in treating Alzheimer's disease in the animal model," Dr. O'Brien said.
